Nicotinamide Riboside is a potent NAD+ precursor that reliably increases cellular NAD+ levels in humans. While it shows significant promise for cardiovascular health and inflammatory markers, evidence for lifespan extension or metabolic improvement in healthy humans remains mixed and largely preliminary.
NR is converted into NAD+ via the NRK pathway, bypassing the rate-limiting NAMPT enzyme. Increased NAD+ availability supports sirtuin activity and mitochondrial oxidative phosphorylation.
